The pathway of myo-inositol degradation in Aerobacter aerogenes. Conversion of 2-deoxy-5-keto-D-gluconic acid to glycolytic intermediates.

چکیده
The product of the first two reactions in the enzymatic degradation of myo-inositol by extracts of Aerobacter aerogORBS, D-2,3-diketo-4-deoxy-epi-inositol, serves as the substrate for another enzyme also present in these extracts. The enzyme was partly purified and could be shown to catalyze, presumably after a preliminary isomerization, the hydrolytic cleavage of the inositol ring. The product of the reaction was isolated and identified by the study of its chemical and physical properties as a 4-deoxy-S-ketohexonic acid.

The Pathway of mpAnosito1 Degradation in Aerobacter aerogenes
The enzymes responsible for the first two steps in the degradation of myo-inositol by Aerobacter aerogenes were identified and isolated from extracts of a “constitutive” mutant, that is, a strain containing high levels of these enzymes even when grown in an inositol-free medium. The enzyme catalyzing the initial step, the dehydrogenation of myo-inositol, is strongly inhibited by its product. It...
متن کاملRegulation of myo-inositol catabolism in Aerobacter aerogenes.
A mutant of Aerobacter aerogenes produces constitutively the series of enzymes that mediates the degradation of myo-inositol and which in the wildtype strain is inducible. When grown on l-histidine, the mutant forms the enzymes at a level approximately three times as high as that seen in the induced wild type. The enzymes appear to be coordinately regulated and are sensitive to catabolite repre...

Department of Microbiology, University of Reading, Whiteknights, PO Box 228, Reading RG6 ZAJ, UK On the basis of enzyme assays, myo-inositol appears to be catabolized via 2-keto-myo-inositol and ~-2,3-diketo-4-deoxy epi-inositol in Rhizobium leguminosarum bv. viciae, as occurs in Klebsiella aerogenes. The first two enzymes of the pathway, myo-inositol dehydrogenase and 2-keto-myo-inositol dehyd...
متن کاملPathway of L-xylose and L-lyxose degradation in Aerobacter aerogenes.
Pen&e utilization in Aerobacter aerogenes has been shown from fermentation studies with Cl%beled pentoses (1, 2) and with cell extracts (3) to involve the conversion of the pentose to n-xylulose 5-phosphate, which is further metabolized via n-sedoheptulose 7-phosphate, n-fructose 6-phosphate, and intermediates of the Embden-Meyerhof pathway. Although Aerobacter aerogenes can grow on all eight o...
متن کاملCarbohydrate Oxidation by Pseudomonas Ruorescens
Glucose utilization by extracts of Pseudomonas jhwrescens involves its oxidation to gluconate and 2-ketogluconate (1, 2) by cytochrome-linked dehydrogenases (1). 2-Ketogluconate is phosphorylated by an inducible kinase, which is distinct from gluconokinase, and 2-keto-6-phosphogluconate, the phosphorylation product, has been isolated and identified (3). The conversion of 2-keto-6-phosphoglucona...
